【摘要】鞘脂在癌细胞的生长和死亡中起着关键作用,越来越成为新型抗癌疗法的主题。鞘脂途径中的几种酶有助于神经酰胺和S1P 的稳态。神经酰胺酶是一种溶酶体半胱氨酸水解酶,可催化神经酰胺转化为脂肪酸和鞘氨醇。在在人类中,已知有五种不同的神经酰胺酶———酸性神经酰胺酶、中性神
经酰胺酶和碱性神经酰胺酶1、2和3,它们由五种不同的基因(分别为ASAH1、ASAH2、ACER1、ACER2和ACER3)编码组成。于神经酰胺的增加和随之而来的S1P 的减少会减少各种癌症的增殖,因此神经酰胺酶可能为抗肿瘤提供新的靶点。文章旨在评估目前神经酰胺酶在鞘脂代谢和肿瘤中可能发挥的潜在作用。
