Beclin-1、LC3B在神经母细胞瘤组织中的表达及意义
摘要
目的:检测神经母细胞瘤及正常淋巴结组织中LC3B与Beclin-1的表达,分析LC3B与Beclin-1的表达在两者中的差异,并进一步分析LC3B 与Beclin-1在神经母细胞瘤中的表达与神经母细胞瘤临床病理特征的关系,探讨其在神经母细胞瘤中的意义。
方法:本研究纳入自2010年10月至2018年12月间就诊于河北医科大学第四医院并经病理诊断为神经母细胞瘤的38例患者及16例儿童肿瘤患者的正常淋巴结组织为研究对象。采用S-P免疫组化方法检测标本中LC3B及Beclin-1的表达,用χ2检验或Fisher精确概率法分析两者表达与临床特点的关系。采用Spearman相关分析法分析LC3B和Beclin-1之间的相关性。
泰国屠杀结果:1、LC3B阳性表达位于细胞浆,Beclin-1阳性表达位于细胞核,LC3B及Beclin-1在神经母细胞瘤表达阳性率高于正常淋巴结组织,差异具有统计学意义(P<0.05)。2、LC3B及Beclin-1在神经母细胞瘤中的阳性表达与病理类型及危险度分组相关,危险度高、病理类型分化差,两者表达阳性率高,
差异均具有统计学意义(P <0.05),与性别、发病年龄、血清NSE、LDH及尿VMA水平等无显著差异(P>0.05)。3、Spearman 相关分析结果显示:LC3B与Beclin-1在神经母细胞瘤中的表达呈正相关(r=0.447, P =0.006)。
木年轮结论:1、自噬相关蛋白LC3B与Beclin-1在神经母细胞瘤中表达明显高于正常淋巴结组织,且与神经母细胞瘤临床危险度分层、分化程度密切相关,提示自噬参与了神经母细胞瘤的发生发展过程。
2、LC3B与Beclin-1在神经母细胞瘤表达成正相关,Beclin-1与LC3B 在神经母细胞瘤自噬过程中可能起协同作用。
3、LC3B与Beclin-1在神经母细胞瘤中阳性表达与临床危险度分层及病理类型密切相关,可协助判断神经母细胞瘤病情程度和预后。

Expression and significance of Beclin-1 and LC3B in the
neuroblastoma
ABSTRACT
Objective: This study aimed to detect the expression of LC3B and Beclin-1 in the neuroblastoma and normal lymph node tissues, and analyzed the differences of LC3B and Beclin-1 between neuroblastoma and normal lymph node tissues , and analyzed the relationship between LC3B and Beclin-1 expressions in neuroblastoma with the clinicopathological features of neuroblastoma in its sufferers.
Methods:This study used data from 38 patients diagnosed with neuroblastoma and 16 patients’ normal lymph nodes seen at The Fourth Hospital of Hebei Medical University between October 2010 and December 2018. The expression of LC3B and Beclin-1 in specimens was detected by immunohistochemical staining. The relationship between the expression levels of LC3B and Beclin-1 and clinical features was analyzed by Chi-square test or Fisher exact probability method. Spearman correlation analysis was used to explore the correlation between LC3B and Beclin-1. P <0.05 was considered statistically significant.
Results:1. The expression of LC3B was located in cytoplasm, and the expression of Beclin-1 was located in nucleus .The expression of LC3B and Beclin-1 in neuroblastoma was significantly higher than that in normal lymph node tissues.2.The positive expression of LC3B and Beclin-1 in neuroblastoma was related to pathological type and risk group.There was no significant difference in
sex, age or tumor markers (NSE, LDH, VMA). 3. Spearman correlation analysis showed that the expression of LC3B and Beclin-1 were positively correlated in the neuroblastoma (r=0.447, P=0.006).
Conclusions: 1. The expression of LC3B and Beclin-1 in neuroblastoma is higher than that in normal tissus,and is related to the pathological type and risk group of the neuroblastoma . LC3B and Beclin-1 may play a role in the
development of the neuroblastoma. 2. The expression of LC3B and Beclin-1 are positively correlated in the neuroblastoma , Beclin-1 and LC3B may play a synergistic role in autophagy process of neuroblastoma. 3. The expression of LC3B and Beclin-1 is related to the pathological type and risk group of the neuroblastoma, which may help to estimate the severity ang prognosis of the neuroblastoma.
