胃癌环状RNA表达谱研究及其功能分析

目的:筛选并鉴定胃癌circRNA表达谱,分析circRNA的生物学功能。
方法:体外培养人低分化胃癌细胞MGC-803、人中分化胃癌细胞SGC-7901、人高分化胃癌细胞NCI-N87和人正常胃上皮细胞GES-1,利用circRNA基因芯片技术检测胃癌细胞和正常胃黏膜上皮细胞中差异表达的circRNAs,利用qRT-PCR技术对部分差异表达的circRNAs进行验证,使用Cytoscape软件构建circRNA-miRNA-mRNA网络图,对下游靶基因进行GO分析和KEGG Pathway分析。
结果:与人正常胃黏膜上皮细胞相比,在不同分化程度的人胃癌细胞中,表达上调的circRNA为79个,表达下调的circRNA为229个。PCR 结果提示hsa_circRNA_103571在正常细胞中明显高于肿瘤细胞,GO分析提示hsa_circRNA_103571间接调控的靶基因在分子功能方面,主要集中在结合蛋白、调节蛋白激酶活性等方面;在细胞组分方面,主要集中在细胞内细胞器方面;在生物学过程方面,主要集中在参与细胞蛋白质修饰等方面。KEGG Pathway分析提示hsa_circRNA_103571可能通过Ras信号通路、FoxO信号通路、Hippo信号通路以及mTOR信号通路参与胃癌发生发展。

结论:在胃癌细胞中,hsa_circRNA_103571是抑癌基因,其机制可能是与hsa-miR-16-5p相互作用,通过Ras信号通路、Hippo信号通路等多条信号通路参与胃癌的发生发展。

Objectives:To screen and identify the expression profile of circular RNA in gastric cancer cells and analyze its biological function.
Methods:Human poorly differentiated gastric cancer cells MGC-803, human moderately differentiated gastric cancer cells SGC-7901,human highly differentiated gastric cancer cells NCI-N87and human normal gastric epithelial cells GES-1are cultured in vitro.The differentially expressed
circRNAs in gastric cancer cells and normal gastric epithelial cells were detected by circRNA gene chip technology.Some differentially expressed circRNAs are validated by qRT-PCR.Cytoscape software is used to construct circRNA-microRNA-RNA network map,GO analysis and KEGG Pathway analysis are performed on downstream target genes.
Results:Compared with normal gastric epithelial cells,79up-regulated circRNAs and229down-regulated circRNAs were expressed in human gastric cancer cells with different differentiation degrees.The results of PCR indicated that hsa_circRNA_103571was significantly higher in normal cells than in cancer cells.GO analysis indicated that the target genes indirectly regulated by hsa_circRNA_103571mainly focused on binding proteins and

regulating the activity of protein kinase,cell components mainly concentrated on organelles,biological processes mainly involved in protein modification. KEGG Pathway analysis suggests that hsa_circRNA_103571may participate in the development of gastric cancer through Ras signaling pathway,FoxO signaling pathway,Hippo signaling pathway and mTOR signaling pathway.
Conclusions:In gastric cancer cells,hsa_circRNA_103571is a tumor suppressor gene,and its mechan
ism may be to interact with hsa-miR-16-5p, and participate in tumor development through multiple signaling pathways such as Ras signaling pathway and Hippo signaling pathway.
