ORM2在肝细胞癌中的表达及功能研究

摘要

肿瘤转移是一个受多种因素调控的复杂过程。Orosomucoid(ORM2)是一种肝细胞合成、分泌的酸性糖蛋白，作为急性反应期蛋白，主要在抗炎、免疫调节以及作为药物运输的载体中起重要的作用。

本研究通过免疫组织化学对原发性肝细胞癌患者临床标本分析发现，ORM2与肝细胞癌的肝内转移以及组织学分级存在负相关。为进一步研究ORM2在肝细胞癌发生发展中的功能，我们构建了ORM2慢病毒表达载体，在SMMC-7721细胞、Li-7细胞与HCC-LY5细胞中过表达ORM2后，在体外抑制肝细胞癌细胞的迁移侵袭能力，在体内可以抑制生长及肝内转移。在Huh7细胞及PLC/PRF/5细胞干扰内源性ORM2的表达可以促进肝细胞癌细胞的迁移侵袭能力。

转录因子CCAAT增强子结合蛋白β(CCAAT/enhancer binding proteinβ，C/EBPβ)是调控ORM2表达的重要转录因子，它有三种变异体，分别为LAP1、LAP2和LIP，但只有LAP1和LAP2能够与ORM2的启动子结合并能上调ORM2的表达。并且我们发现LAP1可以通过上调ORM2进而抑制肝细胞癌细胞的迁移侵袭作用。通过免疫组织化学对原发性肝细胞癌临床样本的分析发现，C/EBPβ与肝细胞癌的组织学分级存在负相关，而且在肝细胞癌组织中C/EBPβ的表达与ORM2的表达存在正相关（r=0.172,p=0.008）。禁欲主义

综上所述，在肝细胞癌细胞中C/EBPβ可以明显上调ORM2的表达；ORM2能够体内外抑制肝细胞癌细胞的迁移侵袭能力，其在肝细胞癌组织中的表达与C/EBPβ的表达密切相关，并与肝细胞癌患者的肝内转移呈负相关。

ABSTRACT

Tumor metastasis is a complex process,and the incidence of metastasis is influenced by many biological factors.Orosomucoid2(ORM2)is an important acid glycoprotein that is mainly biosynthesized and secreted by hepatocytes.As an acute-phase protein,ORM2likely plays important roles in anti-inflammation, immunomodulation and drug delivery.

chineseladyIn this study,we detected and found that ORM2expression in HCC tissues was negatively associated with intrahepatic metastasis and histological grade.Moreover, we established stable ORM2-overexpressing cell lines with SMMC-7721cells,Li-7 cells and HCC-LY5cells,and knockdown the expression of ORM2in Huh7and PLC/PRF/5cell lines.The results showed that the ectopic overexpression of ORM2 decreased HCC cell migration and invasion in vitro and intrahepatic metastasis in vivo, whereas silencing ORM2expression resulted in increased tumor cell migration an

d invasion in vitro.

Furthermore,it was found that the CCAAT/enhancer binding proteinβ(C/EBPβ) upregulated ORM2expression,in which only the LAP1/2C/EBPβisoforms possesse transcription-promoting activity on the ORM2promoter.We also found that LAP1 repressed HCC cell migration and invasion via inducing ORM2expression.The expression of C/EBPβwas found to be positively related to that of ORM2(r=0.172, p=0.008).

Taken together,the expression of C/EBPβprotein was negatively associated with histological grade and positively correlated with ORM2protein expression in HCC tissues.Our findings indicate that ORM2functions as a tumor suppressor and suppresses HCC metastasis through regulation of C/EBPβ.
