摘要 真核细胞核膜上的核孔复合体(nuclear pore com plex,NPC是细胞核内外进行物质交换的主要通道,分子量较小的化合物可自由通过NPC或采取被动扩散的方式进入细胞核,而
分子量为50 kD以上的蛋白质则只能通过主动转运进入细胞核.以这种方式进入细胞核的蛋白质必须在其氨基酸序列上拥有特殊的核定位信号(nuclear localization signal,N LS,以被相应的核转运蛋白(kary opherins识别.核定位信号具有多样性,包括经典核定位信号(classical N LS,cN LS,内输蛋白β2识别的核定位信号(又称PY模体2N LS和其它类型的N LS.每一类N LS具有相似的特征,但并不具有完全保守的氨基酸组成.不同的N LS,往往对应着各不相同的核输入机制.而对同一蛋白质来说,也可能同时拥有几个功能性的N LS.研究核定位信号一方面可以帮助揭示新的大分子物质核转运机制,另一方面也有助于发现一些蛋白质的新功能.本文对常见N LS的分类进行了总
结,并介绍了两种常用的N LS预测软件及鉴定N LS的一般策略.

Abstract The nuclear pore com plex(NPCis in charge of the exchange of macrom olecules between the nuclei and the cytoplasm of eucary otic cells.The small com pounds can get into the nuclei through the NPC freely or by passive diffusion.H owever,those proteins whose m olecular weights are larger than50kD can only get into nuclei by active transport.There must be one or m ore special signals in the amino acid sequences,s o2 called nuclear localization signals,which can be recognized by respective kary opherins,in those proteins.
There are diverse sequences in N LSs,including classical N LS(cN LS,PY2N LS,which can be recognized by im portinβ2,and s ome other N LSs.Though there are similar properties am ong the same family members of N LSs,there is no fully conserved amino acid sequence.Different N LSs often correspond to different mechanisms of nuclear transport,and als o,couple of functional N LSs may exist in one certain protein.The studies on N LS help us not only reveal new nuclear transport mechanism of macrom olecules,but als o discover new functions of proteins.Categ ories of N LSs were summarized by the authors.T w o comm on2used s oftware em ployed in N LS prediction and the strategies used to identify N LS are als o introduced in this review.
