铁转运蛋白编码基因SLC40A1在头颈鳞癌中表达及影响*

530021)摘要目的:探讨SLC40A1在头颈部鳞状细胞癌(HNSCC )中的转录和表达水平,及其与患者临床病理特征之间的关联。方法:基于公开数据库GEO 和TCGA 中HNSCC 的数据,分析SLC40A1在肿瘤组织和正常组织中的转录水平差异。利用On-coLnc 在线数据库研究SLC40A1转录水平与患者总生存率之间的关系。利用免疫组织化学技术验证HNSCC 中铁转运蛋白的表达下调。综合分析SLC40A1的转录和表达水平与患者临床参数之间的关系。结果:基于GEO 的两个数据集和TCGA 数据库分析,均提示SLC40A1基因在HNSCC 中转录下调(P <0.05),其转录水平和患者临床分期相关。生存分析显示,SLC40A1表达低的患者生存时间较短(HR =0.4954,95%CI :0.28~0.89,P =0.0185)。组织芯片免疫组化结果提示,SLC40A1在HNSCC 组织中表达下调(P <0.05)。结论:SLC40A1在HNSCC 中普遍表达下调,且与其不良预后有关;SLC40A1基因的转录和表达失活可能影响肿瘤的发生和演进。

Abstract Objective:To investigate the transcription and expression level of SLC40A1in head and neck squa-mous cell carcinoma (HNSCC)and to discussits relationship with the clinicopathological characteristics of HN-SCCpatients.Methods:Based on the data of HNSCC in the public databases GEO and TCGA,the differences in transcription levels of SLC40A1in tumor and normal tissues were analyzed.The OncoLnc online database was used to study the relationship between SLC40A1transcription level and the overall survival rate of HNSCCpa-tients.Immunohistochemistry was used to verify the down-regulation of iron transporter expression in HNSCC.Comprehensive analysis of the relationship between SLC40A1transcription and expression levels and patient clin-ical parameters.Results:Based on two GEO data sets and TCGA database analysis,it was suggeste
d that the SLC40A1gene was down-regulated in HNSCC (P <0.05),and its transcription level was related to the clinical stage of the HNSCCpatient.Survival analysis showed that patients with low expression of SLC40A1had shorter survival time (HR=0.4954,95%Cl:0.2-0.89,P =0.0185).The results of tissue chip immunohistochemistry sug-gested that the expression of SLC40A1was down-regulated in HNSCC tissues (P <0.05).Conclusion:SLC40A1is generally down-regulated in HNSCC and related to the poor prognosis of HNSCCpatient.Inactivation of the transcription and expression of the SLC40A1gene may affect the occurrence and progression of tumors.Keywords SLC40A1;head and neck squamous cell carcinoma;bioinformatics;molecular marker
