摘要
目的:实验从脑病理组织切片、黑质部位凋亡细胞的形态和数量以及钙离子含量的变化方面来探讨疏筋解毒方对6—羟基多巴胺(6—hydroxydopaminne,6—OHDA)所致帕金森病(Parkinson’s disease,PD)大鼠模型的防治作用,从而揭示疏筋解毒方保护多巴胺(dopamine,DA)能神经元以及对PD作用的可能机制,为中药PD提供一定的实验依据和临床参考。
方法:实验选取Sprague-Dawley(SD)雌雄各半的大鼠,随机分为六组:假手术组、模型组、左旋多巴(L-dopamine,LD)组(LD组)、疏筋解毒方小剂量组(SJS组)、疏筋解毒方中剂量组(SJM组)和疏筋解毒方大剂量组(SJL组)。采用偏侧黑质纹状体6—OHDA立体定位注射术建立PD大鼠模型,分别进行了PD大鼠行为学观察;应用HE染法在普通光学显微镜下观察了脑病理组织切片;应用黑质致密部DNA末端标记法检测了凋亡细胞的形态及数量;应用甲基百里香酚蓝比法检测了黑质纹状体组织钙结合蛋白的含量。
实验结果:
1 疏筋解毒方对PD大鼠黑质组织形态的影响
HE染结果:与其它组相比,SJM组大鼠中脑黑质神经细胞较丰富,排列较密集,形态及体积基本接近正常。
2 疏筋解毒方对PD大鼠中脑黑质神经元凋亡的影响
TUNEL染结果:关于凋亡细胞数目,各组与模型组比较均有显著性差异,P<0.01;而中药组与西药组比较也有显著性差异,P<0.01;且SJM组与其它组比较均有显著性差异,P<0.01。关于阳性目标面密度比较,SJS组范围最大,除假手术组外LD组范围最小。关于阳性目标数密度比较,除模型组外LD组值最大,除假手术组外SJM组值最小。
3 疏筋解毒方对PD大鼠黑质纹状体钙结合蛋白含量的影响
甲基百里香酚蓝比法结果:LD组与模型组无差别;SJS组和SJL组与模型组比较有显著性差别,P<0.01,但与假手术组比较也有差别,P<0.05;而SJM组与模型组有差别,P<0.05,与假手术组无差别。

结论:
1 中剂量的疏筋解毒方可以有效地改善PD大鼠黑质组织的形态,对PD中脑神经元及神经细胞具有保护作用。它可能通过修复受损的神经细胞,恢复细胞产生DA的功能,增加体内DA的含量,从而达到DA与Ach两种神经递质的平衡,从根本上改变PD大鼠体内的生化代谢状态,阻止疾病的进程,对PD大鼠产生一定的脑保护作用。

2 疏筋解毒方可能对细胞氧化应激条件下的凋亡通路具有抑制作用,从而阻抑DA能神经元的变性,其中中剂量的疏筋解毒方抑制黑质细胞凋亡、保护中脑DA神经元的浓度最为合适。

Shujin Jiedu Prescription to Inhibit substantia nigra
apoptosis and Mitigate PD
Abstract
Aim:Through the PD big mouse behavior study the brain pathological tissue slice up observation,examinating the shape and quantity of apoptotic cells and the change of the calbindin content in substantia nigra, the experiment discusses the prevention and curl function of the Shujin J
iedu Prescription to the PD big mouse model established by 6—OHDA,reveals the possible mechanism of the Shujin Jiedu Prescription about how to protect DA neurons and curl PD,then provides certain experiment gist and clinic preference for the Chinese traditional medical treatment of PD.
Methods:The experiment selects Sprague-Dawley(SD)mouse,divides into six groups stochastically:The sham-operation group,the model group,the L-dopamine(LD) group.

1 The impact of the Shujin Jiedu Prescription on PD mouse substantia nigra tissue shape.
HE dye results showed: Compares with other each group of PD mouse’s,the SJM group big mouse’s midbrain substantia nigra neuronal cell are more plenty,more compact in array,more similite with normal mouse’s in shape and volume.
2 The impact of the Shujin Jiedu Prescription on PD mouse midbrain substantia nigra neuronal apoptosis.
TUNEL dye results showed:About the quantity of dead cells, with the model group comparison, each treated group has significantly difference ,P<0.01.With the Western medicine group comparison,each Chinese traditional medicine group has significantly difference,too,P<0.01.And with other  Chinese traditional medicine group comparison, the SJM group has significantly difference,P<0.01.About masculine aim surface density,the SJS group is largest,and the LD group is smallest expect for the sham-operation group. About masculine aim number density, the LD group is largest expect for
the model group,and the SJM group is smallest expect for the sham-operation group.
3 The impact of the Shujin Jiedu Prescription on PD mouse opsoninum corpus striatum in the calbindin content
MTB colorimetry results showed: With the model group comparison, the LD group has no difference,the SJS,SJL group have significantly difference ,P<0.01.But with the sham-operation group comparison, the SJS,SJL group have difference ,too, P<0.05.The SJM group has difference with the model group comparison,P<0.05,and has no difference with the sham-operation group comparison.
Conclusion:
1 The middle dose Shujin Jiedu Prescription may meliorate the PD big mouse's substantia nigra tissue shape effectively,and has protection function to midbrain neuron and neural cell. It possibly through restoring the damaged neural cell, restoring the function of cell to produce DA,increasing DA content in the body,thus reaches the two kinds of neurotransmitter balance
between DA and Ach,alters PD big mouse’s biochemical metabolizing state in the body thoroughly, inhibits the progress of the disease,produces certain protection function to PD big mouse’s brain.
2 Shujin Jiedu Prescription may have inhibiting function to apoptotic pathway on oxidative stress condition,thus inhibit the degeneration of DA neurons.Among all of them, the middle dose Shujin Jiedu Prescription is preferable in its strength to inhibit substantia nigra apoptosis and protect midbrain DA neuron.
3 The middle dose Shujin Jiedu Prescription may be through inhibiting oxidative stress reaction, repairing the structure of cellular velur,improving the mitochondrial function damagement in the cell, inhibiting calcium overload in the cell,reducing the production of free radicel, inhibiting apoptosis, restoring the function of the damaged neuron and neural cell,adjusting hemal acid and alkali degree,thus curls PD roundly.
